It might be a behavior or anything about yourself that you would like to alter. * I would like to change one of my behaviors about myself. The worst quality about me is that I am always late no matter what the case may be. I am not late on purpose, but it seems like something always puts me behind schedule. It drives both my family and friends crazy. They have learned when I say I will be there in thirty minutes, to expect me in about an hour. This is a behavior of mine that I need to work on immediately. . Write a specific goal for making the change, being sure your goal is realistic and fair. Use affirming, motivational language in your goal. For example, ââ¬Å"I will show my best friends that I am paying attention when they talk to me. â⬠* This is something about myself I have needed to change for a long time. It is not something I do intentionally to drive my family and friends crazy. I need to become a little more time oriented, so that I can be where I say I am going to be, at a certain time. I will show my family and friends that I can be on time to their events. My first goal is to sit down with a calendar, and write down already known events and times. This way I will not schedule more than one event at a time. Next, by having a planner, I can add any new events that occur in future. I will be able to check available dates and make sure I have nothing already planned. Lastly, by sticking to my schedule and planning ahead it should reduce the amount of time I am late. I know I always have to expect the unexpected, but at least I have control over the already known about. 3. Observe or think of others who are models for what you want to be. Write down what they do. For example, ââ¬Å"Tracy nods a lot and repeats back what others say so they know she is listening. â⬠* A model person for me to observe and take tips from would be my mother. She will have a whole list of things to accomplish within her day, and gets them all done and on time. She has a monthly day planner that she carries with her at all times. She always makes sure to add important and upcoming events, as well as any of her daily to-dos to the planner. My mom also makes sure not to make more plans than she can fulfill within her day. It seems to work for my mom, so I donââ¬â¢t see why it wouldnââ¬â¢t work for me. . What context or setting is required for you to achieve your goal? For example, ââ¬Å"I will talk with my friends in private, distraction-free settings. â⬠* I will need to follow the schedule I set for myself and not let myself get distracted from my original plans. It might even be good for my family and friends to be a little bit stricter when it co mes to my tardiness. For the most part, this behavior is something I have to train myself on. No one else can do this for me. I have got to learn how to manage my time better, so that I will arrive at events when I am supposed to be there. . Identify potential barriers to the change. Some potential barriers that may cause some problems for my change will be the exact people who request the change of me. I will admit that I do control a lot of my tardiness, but not all of it is my fault. I will be on my way somewhere, when a friend asks me to do something for them. They promise it will only take five minutes. At that point, it is usually over an hour when they are done with me. I have got to learn to tell my family and friends no sometimes, in order to fulfill prior obligations.

Evolutionary Analysis of Culex Species INTRODUCTION: Culex, a mosquito belongs to the phylum Arthopoda, is an important factor causing filariasis [1] St. Louis encephalitis [2]West Nile virus [3] Avion malaria (4). C.quinquefasciatus also called the Southern house mosquito is extensively studied as it transmits crucial diseases [5]. C. pipiens found in the urbans (6) is because of swift urbanization uncanny city growth which eventually resulted in the progression the disease transmittors (7,8,9). Lymphatic filariasis is spread worldwide, affecting 120 million peopleà [10]. They bite on the foot regions of humansà (11) causing skin allergy associated with irritationà (12).Since 1979 there are reports Japanese encephalitis in India, AP and was again reported in 2003à [13]. Known as a cosmopolitan mosquito, acts as vector for protozoan parasites (14 ),filarial worms (15), and for arboviruses(16 ,17). Cx. quinquefasciatus is the prime vector in India causing filariasisà (18) which involve 91% caused by Wuchereria bancroftiCobbold à [19] Hence, therefore, it is very important to eradicate this for the human welfare for which it is very essential to understand them at their molecular levels. The genome sequence of Culex quienquefasciatusà [20]triggered a new hope for the researchers. However, the researchers are still involved in identifying and characterization of these species causing diseasesà (21). The objective of the present article is to access the evolutionary relationship among the culex species with the DNA barcoding and to assess the evolutionary relationship with Tamura 3 Parameter. To accomplish the objective, it is important to use the advanced techniques like the computerized data assessments (22) which could have an immense impact on the health care S. MATERIALS AND METHODS: 2.1 Larvae collection and Characterization: Culex larvae were sampelled from various parts of Hyderabad during the breeding season from different locations, say stagnant water, coconut shells, tires etc. Thus collected larvae were reared and were identified morphologically for Culex quienquefasciatus under the microscope. Microscopic analysis revealed the following characteristics, Short and stout head, yellow long mouth brushes. It was also observed that the abdomen has eight segments, the saddle and the siphon, which is four times longer that its width, securing the larvae to be Culex quinquefasciatus. The larvae were allowed to grow into an adult (27). Further study was performed from the F1 generation of the pure culture after they were identified morphologically for adult. 2.2 DNA EXTRACTION: From the 4th instar larvae, the total DNA was extracted and was then finely ground in 50 à µl of homogenate buffer (28). The homogenate was left in the thermo cycler for 30 minutes at 60oC with a quick addition of 7 à µl of 8M CH3COOK. Incubate the tubes in ice for 30 minutes and centrigugate them for at a maximum speed for 15 minutes. Transfer the supernatant into fresh tubes. To precipitate the DNA, incubate the tubes for 15minutes after adding 100 à µl of 95 % ethanol. DNA pellet was collected after a rapid centrifugation for 15 minutes at maximum speed and decay the supernatant. Air dry the pellet and suspend it in Tris buffer. Store it at 20oC till further experimental procedure is carried. COI Partial sequencing, amplification, Sequencing and Alignment: The isolated DNA was further amplified on PCR by mitochondrial Cytochrome c Oxidase I (COI) gene, which can differentiate between the diversity of taxa . The mitochondrial COI gene of ~ 500 bp was amplified by forward and reverse primers (28) which were developed on the spanning, ~700 bp of Aedes aegypti, Anapheles stepfheni and Cx. Quienquefasciatus. The reaction mixture, 25 à µl, consisting of 10-50 ng of DNA template, 200à µM of dNTPSââ¬â¢ 1U of Taq DNA polymerase, 1X assay buffer and 5p mol of primer was then subjected to amplification for 2 minutes at 94o C initiation, denaturation of DNA template for 35 cycles for 30 seconds at 95 o C, followed by primer aliening -30 seconds at 55 o C, extension- 70 o C for 30 seconds and final extension at 70 o C for 10 minutes. The amplified sequence ,thus, was run on Agarose gel electrophoresis and then the sequencing was perfoemed by Bioserver Biotechnologies Pvt. Ltd. The sequence was submitted to NCBI. The accession number assigned was JX08870 (501bp). Further multiple sequence alignment was performed for partial COI gene sequence of 501bp with the default parameters. Sequence alignment studies elucidate the similarity and differences among different species in India along with other parts of the world. Further, the results of the DNA barcoding were made increasingly vivid with the phylogenetic analysis by the construction of phylogenetic tree. The analysis of the phylogeny was attained by maximium likelihood method (29) with the deletion of gaps and missing data. Bootstrap replication was used to validate the tree. RESULTS AND DISCUSSIONS: Known as the ââ¬ËSouthern House Mosquitoââ¬â¢, Cx. Quienquefasciatus a vector which played a prime role in filariasis and the incidences have seen in different parts of the world. Hence, there is an utmost need to address this problem with priority. In order to snub the prognosis of this vector, it is very essential to take certain control measures. To attain this, the mosquito larvae were collected and grown into an adult. The DNA was isolated and amplified and run on the agarose gel electrophoresis. The agarose wells comprised of sample from different locations along with 200bp marker DNA. The amplicons were seen between 400bp and 600bp of the DNA marker and were then subjected to sequencing. It was noted that the sequence was identified to be novel and was submitted to NCBI-gene bank nucleotide database and the accession number assigned was JX088701. Further, the multiple sequence alignment was performed to understand the evolutionary relationship among the species of the world.. Using the Maximum Likelihood Method, which creates a tree of highest Likelihood from the given data. The Maximum Likelihood tree elucidates that the Culex species of Hyderabad was closely related to the UK species and hence both emerged as out group in the phylogenetic tree. CONCLUSION: The medium sized brown mosquito, Culex quinquefasciatus, predominantly exists all through the tropics acts as a vector causing several parasitic diseases. Usually active at night, it is an opportunistic blood feeder which allows the parasites to use the humans as hosts. It is hence important to address their control with priority. The present article the DNA sequence was successfully isolated, sequenced and was submitted to NCBI-Gen Bank nucleotide database. Further, to understand its evolutionary relationship the phylogenetic analysis was performed. The most important step is to provide clear communication of what the instructions are and your expectations regarding the task. The real reward will be the success of their students and the growth of their knowledge of the world around them. I have observed that becoming a teacher is different from training or studying for any other job. Teachers bring their work home with them, both literally and figuratively. All teachers need to bring home papers to grade and lesson plans to work on; yet, becoming a real teacher means taking the job more seriously. Teachers are mentors who tackle little problems that a particular student faces. Teachers deal with the challenges of making lessons fun, creative, and effective. Being a compassionate, caring teacher is a difficult task because educators only have limited time to really make an impact on their students. Most classes only last about forty minutes each day. I also walked with short, dainty steps, to show how fragile and slight she is. When we sat down, I leaned forward slightly, with my shoulders slightly hunched, to show her vulnerability. Finally we were asked to portray Stella ââ¬â standing upright, slightly tense as if waiti ng to heed to Stanleyââ¬â¢s latest demands ââ¬â and Mitch ââ¬â big, awkward, slightly more hunched and less confident-looking than Stanley. After this task, we were asked to depict Stanley and Blanche as animals. For Stanley, nearly everyone acted like a wolf or a fox ââ¬â strong, territorial, sly, always hunting and watching. For Blanche, I chose to act like a small bird, which would preen and look pretty but would be fidgety and nervous and would fly away when frightened. Our next task was to get into groups and act out scenes taking place before the play begins, which would reveal something interesting about our characters. I was in a group with Emily, and we were told to act out a scene from when Blanche and Stella still lived at Belle Reve. Emily played Stella and I played Blanche. Our scene began with Blanche climbing through the window of the room she shares with Stella, late at night. Stella, who has been studying, admonishes her for being so late and tells her how worried she has been. Blanche, who has been drinking but claims she ââ¬Å"limits herself to oneâ⬠, has just met a young man who reads poetry, who, as we know, will later commit suicide. It is revealed that Stella is the clever sister, who has the potential to make something of herself, and Blanche is the wilder, worldlier sister who relies on her beauty to have fun. This explains why Stella left Belle Reve and had the ability to get a husband and a home of her own, and why Blanc he was left behind, unwilling to grow up and surrender her beauty. Millie and Genna also played Blanche and Stella at Belle Reve. Their scene involved Blanche taking Stella out on the town for the first time. Blanche is very domineering and worldly, and Stella is naà ¯Ã ¿Ã ½ve and nervous about the shortness of her dress. Stella wears white, showing her innocence, and Blanche wears red, showing her corrupted virtue. Jack and Owen played Mitch and Stanley. They gave Mitch a deep personality, having him hint about wanting to settle down with a wife and worrying about his mother. They also showed Stanleyââ¬â¢s love for Stella, as he talks about her in a very loving and tender way to Mitch. Next Felix and Flick, playing Stanley and Stella, showed how Stella was very like Blanche when she first met Stanley, unused to this more working class environment, yet being excited by Stanleyââ¬â¢s attentions. Finally, Angelika and Hugo, playing Stella and Stanley, show Stella as being naà ¯Ã ¿Ã ½ve and Stanley being mysterious and inquisitive, trying t o figure her out. In our next task we did Hot Seating. One by one, we were asked questions in character about our views on different events in the play. I chose to be Eunice, because although she is not a main character in the play, she is good friends with Stella and her husband is friends with Stanley, and she lives in the flat above them, so she would have a very clear idea of what life would be like for them. I chose to give her the opinion that Stanleyââ¬â¢s hitting Stella isnââ¬â¢t good, but is acceptable, and is just a part of marriage that cannot be avoided. I was particularly impressed by Emilyââ¬â¢s portrayal of Stella after having her sister taken away. She made it clear that Stella was heartbroken, and although she claimed to believe Stanley, she let on that she had no choice but to believe him. I also liked Alex as Blanche, gushing and acting pleasant and sweet to try and cover up her past, but when difficult questions were brought up her act fell and she would refuse to talk abo ut it. Our final task was to act out the scene where Blanche has been stood up by Mitch on her birthday and use freeze-frames to mark the most important moments in that the scene. I was in a group with Felix and Alex. Felix played Stanley, Alex played Blanche and I played Stella. The moments we marked were: Stanley throwing plates off the table; Stella and Stanley on the porch with Blanche on the phone, trying to reach Mitch; Stanley giving Blanche a ticket back to Laurel; and Stella going into labour. For each of our important moments, we froze in position for four seconds, before doing an action and moving onto the next moment. There were subtle differences between each groupââ¬â¢s piece; instead of using an action, as we did in ours, Millie, Flick and Jack used an important line of text in each of their moments, which I found very effective. Although I found these tasks challenging (especially as I knew we were on camera), I enjoyed interpreting different characters in new ways. I also worked with different people to whom I normally would, which was a good experience and made me enthusiastic and eager to work with them again in future lessons.
